Epigenetic Investigation of Hippo Pathway in Bone, Breast and Prostate cancer cell lines

Epigenetic of hippo pathway genes play crucial roles of signalling pathways in regulating organ size, cell proliferation and cell death in human. Our study focused on identifying differential methyla-tion patterns in bone, breast and prostate cancer cell lines. We performed methylation analysis of the CpG island promoter in these cells by methylation-specific PCR. Our data indicates that methylation of this pathway vary between these cells. No or low methylation frequency was detected in both bone and prostate compared with breast cancer cells. In MST1 and MST2 analysis, 1 out of 3 bone cancer cells showed methylation of the MST1 and MST2 and 3 out of 6 breast cancer cells showed methyla-tion of MST1 and MST2. In case of prostate cancer cells, 1 out of 4 these cell lines showed methylation of the MST1 without detection of any methylation in MST2. In SAV1, 3 breast cells showed methylation was detected compared with 2 of prostate cells and non of bone cancer cells showed any methylation. In LATS1 and LATS2, only 2 breast cells showed methylation of LAST1 compared with 1 of bone cells and non of prostate cancer cells showed any methylation. LATS2 methylation frequency was non detected in all cells. In summary, it is important to understand the epigenetic mechanisms of hippo pathway and to identify epigenetic variations to differentiate between the different cell lines within the same cancer type.
